Use | 2-chloro-4-methoxypyrimidine is an organic intermediate, which can be used as a raw material with 2, 4-dichloropyrimidine It is obtained after nucleophilic substitution with sodium methoxide or potassium methoxide. It can be used as a pharmaceutical intermediate for the preparation of HIV prevention drugs and inhibitors of Bruton's tyrosine kinase. |
Preparation | Add NaOMe(2.84g,52.5mmol) in MeOH(20mL) dropwise to a solution of 2, 4-dichloropyrimidine (7.5g,50mmol) in MeOH(20mL) at 0°C. The reaction mixture was stirred at 0°C for 2h. The reaction mixture is concentrated in a vacuum to obtain a crude product. Pour the crude product into 150mL of water. The aqueous phase was extracted with EtOAc(100mL). The organic layer was dried with Na2SO4 and concentrated in vacuum to give 2-chloro-4-methoxypyrimidine in a pale yellow solid (5.9g, yield: 82%). |
